Description | OriginallydiscoveredasoneofthreegenesamplifiedondoubleminutechromosomesinatumOrigenicderivativeofNIH3T3cells(1),MDM2waslatershowntopossessoncogenicpotentialwhenoverexpressed(2,3).High-levelexpressionofMDM2hasalsobeenshowntoconfertumorigenicpotentialuponnontransformedrodentfibroblastsinathymicnudemice(2,3).MDM2canimmortalizeratembryofibroblastsandcancooperatewithactivatedRAStotransformthesecells(3).ElevatedlevelsofMDM2proteinhavebeenfoundinavarietyofhumantumors,mostnotablyinsofttissuesarcomaswhereupto30%ofprimarytumorscontainmultiplecopiesoftheMDM2gene(4).OnemechanismbywhichMDM2overexpressionpromotestumordevelopmentisthroughitsABIlitytobindtothep53tumorsuppressor,therebyblockingthetransactivation(5-7),cellcyclearrest(8),andapoptoticfunctionsofp53(9).MDM2caninhibitp53activityinanumberofwaysincludingpreventingp53fromrecruitingTAFs,promotingnuclearexport,inhibitingp53acetylation,andperhapsmostimportantlybyvirtueofitsfunctionasanE3ubiquitinligasewithspecificityfor,amongothers,p53(reviewedin10).Inadditiontoregulatingp53levelsbytargetingp53forproteasomaldegradationMDM2alsotransfersubiquitintoitself,MDMX,theß2adrenergicreceptor,glucocorticoidreceptor,TIP60,andPCAF(10). |
