Description | Nuclearreceptorsformthelargestknownfamilyoftranscriptionfactorsandhaveacrucialroleinnearlyallaspectsofvertebratedevelopmentandadultphysiologybytransducingtheeffectsofhormonesintotranscriptionalresponses(1).Thefamilyisdefinedbytwodomains:(a)thecentral,highlyconserved,DNA-bindingdomain(DBD)ofapproximately66aminoacids,and(b)theC-terminal,structurallyconserved,ligand-bindingdomain(LBD)ofapproximately250aminoacids(2,3).Theamino-terminalregionsareleastconservedamongnuclearreceptorsequences.ThisdomainishighlydivergentbetweenTRαandTRβisoforms,whichsuggestsdifferentialrolesintranscriptionalregulation.Inaddition,alternativesplicingoftheTRαgenegeneratestwoisoforms,TRα1andTRα2withcompletelydifferentamino-terminaldomains(4).UnligandedTRinhibitstheformationofafunctionalpre-initiationcomplex,throughdirectinteractionwithTBPandtranscriptionfactorIIB(5-7).Inaddition,intheabsenceofligand,TRhasbeenshowntorepresstranscriptionthroughrecruitmentofacorepressorcomplex,whichalsoincludesSin3Aandhistonedeacetylase(8,9).Ligandbindingreleasesthecorepressorcomplexandrecruitsacoactivatorcomplexthatincludesmultiplehistoneacetyltransferases,includingasteroidreceptorfamilycoactivator,p300/CREB-bindingprotein–associatedfactor(PCAF),andCREBbindingprotein(CBP)(10-14). |
