Description | Theuniversalderegulationofc-Mycgeneexpressionintumorcellssuggeststhatthisoncogenerepresentsanattractivetargetforcancertherapeuticpurposes.Thec-Mycpromoterintegratesdiversemitogenicsignallingcascades,whichareconstitutivelyactivatedintumorcells,andtranslatesthemintoexpressionofthec-Myctranscriptionfactor,whichpromotescellproliferation,growth,differentiation,andapoptosisbyregulatingtheexpressionofnumeroustargetgenes(1-3).ThestructuralandbiochemicalfeaturesoftheMYCfamily(MYC,N-MYC,andL-MYC)markthemasdirectregulatorsofgeneexpression.Asbasichelix-loop-helixleucinezipperproteins(bHLH-ZIP),theMYCsacquirethecapacitytobindtheDNAsequenceCACGTG(E-box)whendimerizedwithMAX(anotherbHLH-ZIP,4,5).Ahead-to-tailpairofMYC-MAXdimersmay,inturn,formaheterotetramercapableofbridgingdistantE-boxes(6).AmongthebroadlydistributedpositiveenforcersofMYCactionthatareoftenrecruitedtotargetgenesarechromatinremodeling(SWI/SNFrelatives)andmodifyingcomplexes(TRAPP/GCN5andrelatives);thesecomplexesmobilizenucleosomesandacetylatehistonesand/orothertargetstoactivategeneexpression(4,7,8).MYCbindsTBPalonganauxiliarypathwaytocontrolgeneexpression.MADandMNTgenerallyopposeMYCactionbyenlistinghistonedeacetylasecomplexes.Besidesactingatthelevelofchromatin,MYCmayalsooperateatlaterstagesofthetranscriptioncycle,afterpre-initiationcomplexformation(9).Inadditiontousinggenericchromatincomplexestoup-ordown-regulatetranscription,theMYCnetworkalsoconscriptsindividualfactorstomodifyexpressionlocallyonanadhocbasis.Forexample,YY1,AP2,MIZ1,SP1,BRCA1,andotherproteinsinteractdirectlywithMYC,andsomaydirectlymodifytheoutputoftheMYCnetwork(10,11) |
